Effect of Panax notogino side on the hypoxia hypercapnia induced pulmonary vasoconstriction

Objective:To investigate the effect of Panax notogino side(PNS) on the main pulmonary artery rings and the underlying mechanisms involving in the role of endothelial cell. Methods: The model of isolated rat pulmonary artery rings in organ baths was performed. Under normoxia and hypoxia hypercapnia conditions, the effect of accumulated PNS on pulmonary artery rings which were pre-constricted by KC1 or norepinephrine(NE) was observed. And the endothelium was removed in part of these pulmonary artery rings. Result: ①Not only under normoxia conditions but also hypoxia hypercapnia conditions, there was concentration-dependent relaxation in pulmonary artery rings pre-constricted by KC(160 mmol/L), with or without endothelium. Similar results occurred in those pulmonary artery rings which were pre-constricted by NE(10-5 mol/L ()P0.05). ②The relaxation effect of PNS on pulmonary artery rings was enhanced under hypoxia hypercapnia conditions. There was statistically significant relaxation in every hypoxia hypercapnia group compared with normoxia group(P0.05 or P0.01). ③ Under normoxia conditions, different vaso-dilation did not occur between endothelium-denuded pulmonary artery rings and endothelium-integrated ones(P0.05). But under hypoxia hypercapnia conditions, PNS caused dramatically relaxation in endothelium-denuded pulmonary artery rings rather than endothelium-integrated ones(P0.05 or P0.01). Conclusion: Under hypoxia hypercapnia conditions, PNS can relax the rat pulmonary rings. Furthermore, in endothelium-denuded groups, dramatically relaxation caused by PNS may result from the depletion of endothelium mediation.
