Endothelial function of aorta in twokidney,one clip renal hypertensive rats

bjective To investigate endothelial function of the aorta in rats with 2K1C renal hypertension.Methods Experiments were performed on aortic ring segments isolated from 2K1C rats 4 weeks after clipping,changes in isometic tension were recorded,and aortic cGMP levels were tested.Results At the 4th week after renal artery clipping,the level of aortic cGMP content was significantly lowered.In isolated aotric ring segments,acetylcholineinduced dilation were suppressed,phenylephrine induced contractile response were markedly enhanced.These changes vanished in 2K1C rats treated with captopril.In contrast,sodium nitroprusside induced maximal dilation did not differ significantly among the three groups.Conclusion The results suggest that the increase in angiotensin Ⅱ level and the deficiency of nitric oxide production may contribute to the vascular endothelial dysfunction in rats with 2K1C renal hypertension. 
