Effect of long-term administration of glucocorticoid on expression level of OPG/RANKL mRNAs in bone tissues of femoral head of rats

Objective To investigate the effect of long-term administration of glucocorticoid on expression level of osteoprotegerin (OPG) and receptor activator of nuclear factor kappa B ligand (RANKL) mRNAs in bone tissues of femoral head of rats, and discuss its mechanism of action on glucocorticoid-induced necrosis of femoral head. Methods Forty adult SD rats were randomly divided into 4 groups: low-dose glucocorticoid group (LD), medium-dose glucocorticoid group (MD), high-dose glucocorticoid group (HD) and control group. The LD, MD and HD groups were treated by intramuscular injection of prednisolone 7, 12.5 and 25mg/kg body weight, respectively, and the control group was only treated by the same volume of sodium chloride. After 4 weeks' intervention, osteonecrosis of left femoral heads was detected by HE staining, the right femoral head bone tissue total RNA was extracted and the expression level of OPG and RANKL mRNAs was examined by real-time quantitative polymerase chain reaction. Results ① The necrosis rate of femoral head was 30%(3/10), 66.67%(6/9), 83.33%(5/6), and 0(0/10) in LD, MD, HD and control groups. ② The level of OPG mRNA was 1.12±0.17, 0.96±0.16, 0.83±0.18 and 1.60±0.49 in LD, MD, HD and control groups. The level of RANKL mRNA was 2.48±0.81, 3.33±1.04, 4.51±1.33 and 1.58±0.42 in LD, MD, HD and control groups. Conclusion The glucocorticoid-induced necrosis of femoral head may be related to the expression level of OPG/RANKL mRNAs regulated by glucocorticoid.
