Effects of matrine on proliferation of human ovarian cancer cells and the action mechanism

Objective To study the effects of matrine on proliferation of human ovarian cancer OV2008 cells in vitro cell culture and the action mechanism.Methods 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide(MTT) colorimetric assay was used to examine the effects of matrine at different concentrations(0,0.125,0.25,0.5,1.0,2.0 and 4.0 mg/mL) and different time points(24,48 and 72 h) on proliferation of OV2008 cells.Flow cytometry(FCM) was used to analyze cell cycle,apoptosis rate as well as expression levels of Fas and Bcl-2 proteins of OV2008 cells treated with matrine at different concentrations for 48 h. Results The results showed that compared with the control group,matrine could significantly inhibit the proliferation of OV2008 cells in time-and dose-dependant manners(P0.05 or P0.01).Cell cycle analysis indicated that the percentage of OV2008 cells was increased in G0/G1 phase and decreased in S phase with the concentration of matrine increased;there was no obvious change in G2/M phase.The results also indicated that matrine could induce apoptosis of OV2008 cells and the apoptosis rate was increased with the increase of matrine concentration compared with the control group(P0.01).Matrine also up-regulated the expression of Fas protein(P0.05 or P0.01) and down-regulated the expression of Bcl-2 protein(P0.01).Conclusion Matrine can effectively inhibit the proliferation of OV2008 cells in time-and dose-dependent manners.The inhibition may be related to the arrest of cell cycle and induction of apoptosis.The matrine-induced apoptosis might be related to the up-regulation of Fas protein and down-regulation of Bcl-2 protein.
