HE Tie-guang~(1,2,3),YANG Li-tao~1,LI Yang-rui~3,SU Jiang~1, WANG Can-qin~2 and WEI Zhu-yang~4 (1.College of Agronomy,Guangxi Univ.,Nanning 530005,China;2.Institute of Biotechnology,Guangxi Academy of Agricultural Sciences,Nanning 530007,China;3.Guangxi Crop Genetic Improvement and Biotechnology Lab,Nanning 530007,China;4.Guangxi Yangping Herba Dendrobii Scientific Research Institute,Yulin 537000,China)
Conditions of the extraction and purification of polysaccharides from suspension-cultured protocorms of Dendrobium candidum were optimized by orthogonal experiment. The results showed that the optimum technology of isolation is to extract about 3 times and 2 hours each time in 20 times water at 80℃,to precipitate polysaccharides with 80% ethanol, and to remove proteins when chloroform/n-butanol is 1∶0.4,the sample/ chloroform-n-butanol is 1∶0.35 and the abstracting time is 5 min.
