Analyses on mtDNA D-loop Genetic Diversity in Guanzhong Horse

The genetic diversity and phylogenetic evolution of 247 bp mitochondrial DNA(mtDNA) D-loop sequences of 27 Guanzhong horse samples were analyzed by sequencing and bioinformation techniques.The results revealed 9 mitochondrial haplotypes with 19 polymorphic sites,covering 7.69% of the analyzed length of the 247 bp D-loop sequences.Among the polymorphic sites,there were all transitions.The nucleotide diversity(π value) and haplotype diversity(H) estimated were 0.0219±0.0076 and 0.843±0.041,respectively,showing high mitochondrial genetic diversity in Guanzhong horse breed.The constructed Neighbor-Joining molecular phylogenetic tree based on the 9 mtDNA D-loop haplotypes,indicated that lineages A,C,D,F and G were detected which showed multiple maternal origins in Guanzhong horse breed.
