SRAP Analysis of Genetic Diversity of Sclerotinia sclerotiorum in Shaanxi Province

We analyzed genetic diversity of 144 Sclerotinia sclerotiorum isolates from different areas and 18 isolates from different hosts in Shaanxi province using sequence-related amplified polymorphism(SRAP)markers.Nine primer pairs were screened out from 150 SRAP primer combinations based on their polymorphism and stable amplification.In this study,76 polymorphic bands were produced by the amplification of 144 isolates,which was 88.4% of the total bands,and the similarity coefficient was from 0.346 to 0.936.Meanwhile,with the amplification of 18 strains from different hosts,we got 62 polymorphic bands which made up 84.9% of the total bands,and the similarity coefficient was from 0.358 to 0.976.The UPGMA(Unweighted Pair Group Method With Arithmetic Mean)cluster analysis showed that the dendrogram consisted of four groups when the genetic similarity coefficient of 144 strains in different regions was 0.658.Group Ⅰ consisted of counties mainly in Hanzhong,while Group Ⅳ consisted of 2 counties in Ankang.Group Ⅱ and Ⅲ are groups from mixed regions.Meanwhile,the dendrogram consisted of three groups when the genetic similarity coefficient of 18 strains in different hosts was 0.788,and 83.3% of them were in groupⅠ consisting of all host types.The dendrogram consisted of three groups when the genetic similarity coefficient of 38 strains of different pathogenicity was 0.730.They are relatively weak pathogenicity isolates in group A,combined pathogenicity in B and C.Results showed a rich SRAP polymorphism among the population of S.sclerotiorum in Shaanxi province but had no significant correlation with genetic diversity and geographical location,nor did the isolates in different host origin and pathogenicity.
