Effects of Different Drought Stresses on Growth and Enzyme Activities Related to Polysaccharide Synthesis of Polyporus umbellatus Mycelium

To explore the effect of the drought stress on the growth and the accumulation of polysaccharides of Polyporus umbellatus mycelium,with different mass concentrations of PEG-6000(0 g/L, 50 g/L, 100 g/L, 150 g/L, 200 g/L and 250 g/L),the strain of P.umbellatus was treated by PEG-6000 to simulate drought stress. Through liquid culture, the growth index and enzyme activities related to polysaccharide synthesis of mycelium were evaluated. The results indicated that the biomass, enzyme activities and polysaccharide mass fraction of P.umbellatus were the highest in the concentration of PEG-6000(100 g/L). Meanwhile, the biomass, HK activity, GPI activity, α-PGM activity and polysaccharide mass fraction significantly increased by 60.40%, 27.88%, 19.89%, 28.73%, 106.00%, respectively, compared to the CK group. When the mass concentration of PEG-6000 was exceeded 150 g/L, the biomass, POD activity, SOD activity, MDA molality, soluble proteins mass fraction and polysaccharide mass fraction of mycelium significantly decreased with the increase of drought stress. Therefore, moderate drought stress are conducive to growth and polysaccharide synthesis of P.umbellatus.
