Simultaneous determination of 3 constituents in Anchusa italica Retz. by HPLC under double-wavelength

Objective To establish an HPLC method for the simultaneous determination of 3 constituents in Anchusa italica Retz.under double-wavelength.MethodsπNAP COSMOSIL C18 column(250 mm×4.6 mm,5μm)was used with the mobile phase of acetonitrile(A)-2 mL·L~(-1) phosphoric acid(B)by gradient elution.The flow rate was 1.0 mL·min-1.The column temperature was30 ℃.The detection wavelength were 266 nm for astragaline and 341 nm for caffeic acid and rutin.Results The method was developed with good linearity over the range of 0.348 8-22.32(r=0.999 9),1.813-116.0(r=0.999 9)and 0.241 3-15.44μg·mL~(-1)(r=0.999 8)of caffeic acid,rutin and astragaline,respectively.The average recoveries of caffeic acid,rutin and astragaline were99.6%(RSD=1.28%),101.1%(RSD=0.66%)and 99.8%(RSD=3.75%),respectively.The 3 samples average contents of caffeic acid,rutin and astragaline were 84.27±2.19,720.77±62.85 and 63.63±4.33μg·g-1,respectively.Conclusion The method is simple,reproducible and precise.The results provide experiment basis for the quality control of Anchusa italica Retz..
