Application of NMR Technique for Assaying Oil Content in Rapeseed

The technique of nuclear magnetic resonance (NMR) was employed to determinethe oil content in rapeseed. Rapeseed oil samples, ranging from 0.19 to 29.48g of oilin CCl_4. were scanned by NMR Analyser, Newport MK Ⅲ A. A high positive corre-lation (r=0.9994) was obtained. The smallest seed amount required was 0.5g. NMRand Soxhlet determinations were compated on 50 repassed samples and coefficientof correlation r=0.9823. The moisture in rapeseeds contributed to the NMR signal, and therefore oilcontent was overestimated. When the moisture in rapeseeds was more than 4%, theanalytic error was linearly increased, at a rate of 0.18%(apparent oil content) one percent moisture in seeds. The accuracy and precision of analysis also varied with thesample and surrounding temperature. The error contributed by surrounding tempera-ture was --0.22%/℃, and from --0.10% to --0.16%/℃ by sample temperature. The methods of drying rapeseeds at 110℃ and 45℃ were recommended to indus-trial, commercial users and breeders. The saturation on rapeseed sample was found at150 μA of the R. F. level. The highest ratio of NMR signal to noise could reach to180 by adjusting the A. F. gain to give a readout to about 100. Six calibrationmethods were suggested to calculate the oil content in rapeseeds.
