Phylogenetic Relationships of Saccharum and Related Species Inferred from Sequence Analysis of the nrDNA ITS Region

The internal transcribed spacer(ITS) regions of nuclear ribosomal DNA from 14 species of Saccharum and related genera were sequenced. The regions ranged in length from 589 to 591bp, with ITS1(205~208bp)slightly shorter than ITS2(216~220bp). Of the 591bp aligned positions, there were 140 variable sites(64 in ITS1, 68 in ITS2, 8 in 5.8S region), and 60 informative sites(33 in ITS1, 24 in ITS2, 3 in 5.8S region). The neighbor joining tree based on the ITS sequences analysis with P. schumach as outgroup indicated that Zhejiang fruit sugarcane should belong to S. officinarum. N. fallax and E. fulvus, N. porphyrocoma and E. rockii should be placed in a same genus. Arundinaceum has different variable sites from other species, it is an independent clade isolated from Saccharum and Erianthus . The present analysis demonstrated that Arundinaceum should not belong to Saccharum , which is consistent with the results from morphology and cytology.
