Protective function of Huanglian Jiedu Tang on Rats after focal cerebral ischemic reperfusion and its influence on caspase-3 expression

Objective It is to observe the protective function of Huanglian Jiedu Tang(HLJDT) on neurocyte apoptosis of Rats after focal cerebral ischemic reperfusion and its influence on caspase-3 expression.Methods 104 Wistar male Rats were randomly divided into sham operation group,operation control group,HLJDT group and positive control group.The latter three groups were further subdivided into one hour,24 hours,48 hours and 7 days subgroups according to different reperfusion time.Cerebral ischemia reperfusion model injury was made manufacture with thread fastening method.The Rats models were done decapitation and taken brain tissue after ischemia for two hours and reperfusion for 1 hour,24 hours,48 hours and 7 days respectively.The infarct site volume,apoptosis cell number and caspase-3 expression were observed and determined with TUNEL staining and SP immunohistochemistry method.Results Sham operation group had no infarction site,apoptosis and caspause-3 positive expression.Compared with that in operation control group,the infarct site volume significantly decreased size,apoptosis cell number decreased and caspase-3 expression weakened in HLJDT group(all P0.05).Conclusion HLJDT can inhibit brain cell apoptosis through downing regulation caspase-3 expression and has protective role on brain.
