Ganoderma lucidum polysaccharides' intervention on the aged endothelial cell induced by angiotensin Ⅱ

Objective It is to investigate the effect of Ganoderma lucidum polysaccharides(GLP) on protecting human umbilical vein endothelial cells(HUVECs) and inhibiting cell senescence.Methods Aging model of HUVECs was established by AngⅡ.HUVECs were cultured in vitro,HUVECs were intervened by AngⅡand divided into three groups: the control group,AngⅡ group,GLP group(given GLP 100 mg/L at 1 h before stimulation with AngⅡ).The cell viability was observed by CCK-8,β-galactosidase activity was measured with immunocytochemical method and cell proliferation activity was measured with flowcytometry.Results Compared with the control cells,the cells positive for β-gal staining was significantly increased in AngII group,cell cycle arrest at G0/G1 phase with decreased S-phase cell percentage and cell viability(P0.01),which indicated that HUVECs senescence were successfully induced by AngII,while the above changes were smaller in GLP intervention group than that in AngII group.Conclusion GLP can inhibit HUVECs senescence,and protect vascular endothelium.
