Influence of androgen receptor expression in osteoblasts on bone metabolism of male rats

AIM: To study the changing regulation of androgen receptor (AR) expression level in the osteoblast of male rats, so as to further investigate the mechanism of androgen in osteoporosis by detecting the change and regulation of serum concentration of testosterone in male rats. METHODS: The study was finished in the Experimental Animal Center, Second Affiliated Hospital of Harbin Medical University from June 2003 to July 2004. The serum concentration of testosterone in male rats was determined with radioimmunoassay (RIA), the osteoblasts were isolated, purified and cultured with sequential enzyme digestion and repetitive stick-to-wall method, and then the osteoblasts were evaluated. The AR in osteoblasts of different months old Wistar rats were determined with the semi-quantitative Western blot method. RESULTS: ①Lower AR expressions were observed in 1 and 2-month-old groups, which were 87.70±2.68 and 90.74±2.06 respectively (staining). The AR expression began to increase in the 4-month-old group, it was obviously increased from the 6th month, and reached the peak levels in the 10, 12 and 14-month-old groups (142.9±4.73,154.2±6.67,190.6±5.80) (staining), while the AR expression showed a decreasing tendency from the 16th month. ②The serum concentration of testosterone showed regular distribution in male rats of different months old. ③There was obvious correlation between serum testosterone and AR expressions (R=0.167 5, P 0.01). CONCLUSION: ①The distribution of AR expression is regular in rats of different months old, and the AR expression is in coincidence with the level of androgen. ②Androgen combines with AR in osteoblasts directly, or turns into dihydrotestosterone (DHT) with the function of 5-alpha reductase, and then combines with AR, so as to regulate a series of function of osteoblasts, which is the main function way of androgen in maintaining the bone stability in vivo.
