Effects of Gypenosides on the stress ability of rats

AIM: To observe the effect of gypenosides on the stress ability of rats under acute stress of different types, and compare the effects with those of ginsenoside.METHODS: The experiment was carried out in the Institute of Biological Engineer, Yichun University between July and August 2004. Ninety Kunming mice and 30 SD rats were used. ② The hypoxia model under normal pressure: Thirty mice were randomly divided into 3 groups: saline group (received the same cubage normal saline), gypenosides group and ginsenosides group (received gypenosides and ginsenosides 100 mg/kg respectively), and the mice were treated for 3 continuous days. At 50 minutes after the last administration, the mice were enclosed into the jars with milling ora of 250 mL containing 10 g limestone, 1 mice in each jar, the jars was made into hypoxia environment, and the last breath was taken as index of death, the survival time of the mice under hypoxia was observed and recorded. ③ The hypoxia model by the effect of isoprenaline: The methods of grouping and administration were same as those mentioned above. The mice were given intraperitoneal injection of isoprenaline (15 mg/kg) at 35 minutes after the last administration, the survival time of the mice was observed and recorded as above after 25 minutes. The models of forced swimming: The methods of grouping and administration were same as those mentioned above. The mice were forced to swim in a water tank of 25 ℃ and 35 cm deep at 50 minutes after the last administration until got down to the bottom of the water tank, the time from swimming to sinking at the bottom of the water tank were observed and recorded. The models of acute myocardial ischemia: Thirty SD rats were divided into 3 groups randomly: saline group (received saline 0.1 mL for each one), gypenosides group (received 30 mg/kg gypenosides), and ginsenosides group (received 30 mg/kg ginsenosides), 10 rats in each group. Cardiogram was recorded after 15 minutes. Then the rats were given injection of 0.5 μg/kg hypophysin (100 μg/L) via sublingual vein to induce acute myocardial ischemia. The change of cardiogram was recorded immediately. It was positive if cardiogram showed as below: ST section was 0.1 mV higher than before experiment; T wave was low and flat; T wave was double direction, T wave showed inversion, heartbeat was irregular. Otherwise it was negative. The group with more negative showed that the drug had powerful protective effect on myocardial ischemia. ⑤ The measurement data were compared with analysis of results, the F test and q test were applied. The enumeration data were expressed as percentage, the χ2 test was performed. RESULTS: All the 90 Kunming mice and 30 SD rats were involved in the analysis of results. ① The results of hypoxia experiment under normal pressure: The survival times in the gypenosides group and ginsenosides group were obviously longer than that in the saline group (P 0.05). ② The results of hypoxia experiment under normal pressure affected by injecting isoprenaline: Both gypenosides and ginsenosides had no obvious effect on the ability of acute stress of mice which were injected with isoprenaline (P 0.05). There was no obvious difference in the survival time among the groups (P 0.05). ③ The results of forced swimming experiment under acute stress: The average swimming time among the groups was not obviously different (P 0.05). But both gypenosides and ginsenosides could extend the forced swimming time, the rate of extending swimming time was 59% and 57%, it was showed that gypenosides and ginsenosides had the tendency of anti-tiredness. ④ The effect of gypenosides and ginsenosides on acute myocardial ischemia in rats: Both gypenosides and ginsenosides could reduce the occurrence of changing rate of ischemic cardiogram (P 0.01, 0.05), it was showed that both gypenosides and ginsenosides had partly effect on myocardial ischemia led by hypophysin, and both of them could protect rats from myocardial ischemia. The negative rates of ischemia in the gypenosides and ginsenosides groups were obviously higher than that in the saline group (P 0.05,0.01).CONCLUSION: Gypenosides can obviously strengthen the ability under acute stress as hypoxia under normal pressure, hypoxia under the effect of isoprenaline and forcing swimming ability under acute stress, it also can strengthen the rats' acute stress under myocardial ischemia, which are similar to those of ginsenosides.
