Bone-marrow mononuclear cells plus bone xenograft for repairing femoral head necrosis

AIM: To observe the repair effect of bone-marrow mononuclear cells combined with bone xenograft on femoral head necrosis. METHODS: The experiment was performed in the surgical laboratory of Department of Orthopedics, First Affiliated Hospital of Liaoning Medical University from April to October 2006. The models of bilateral femoral head necrosis were established in 15 New Zealand rabbits with the liquid nitrogen frozen method after modified by Yang, in which only the articular capsule was cut open, but not the round ligament of femur, and the femoral head was subjected to liquid nitrogen frozen without dislocation. One side of hip joint served as experimental group and the other side as the control. The experimental group was implanted with bone-marrow mononuclear cells mixed with bone xenograft after pressure, and the other with bone xenograft. Radiographical, Masson trichrome staining, and new bone volume percentage analysis were carried out at 2, 4 and 8 weeks after operation to observe the recovery condition of necrotic femoral head.RESULTS: All the 15 rabbits were involved in the result analysis. X-ray examination and Masson trichrome staining results indicated that compared with the control group, the recovery condition of the necrotic femoral head in the experimental group was better. At 2, 4 and 8 weeks after operation, the regenerating trabecular volume percentages were higher than the control group significantly (25.49±0.60)%, (24.44±0.76)%; (32.45±0.37)%, (31.08±0.38)%; (37.93±0.40)%, (36.30±0.74)%, t =2.901, 4.936, 6.627, P 0.05.CONCLUSION: The bone-marrow mononuclear cells combined with bone xenograft could well repair the necrotic femoral head.
