Correlation between content of brain dopamine and the power of brain 11 mHz infra-slow wave spectrum in rats

BACKGROUND:Changes of cerebral neurotransmitters is an important contents in neuroscience,however,it is lack of neurotransmitters detection means at the noninvasive condition. OBJECTIVE:To explore the corresponding relation between powers of brain 11 mHz infra-slow wave spectrum and content of dopamine under the conditions that the brain transmitter content lower than or equal to the normal level. DESIGN,TIME AND SETTING:A randomized,controlled animal experiment was performed at the Beijing Institute for Neuroscience,the Capital Medical University from July 2006 to February 2008. MATERIALS:One hundred female,clean degree,Sprague Dawley rats weighted 200-250 g were divided into model (n=40),control (n=30) and normal (n=30) groups according to random table method. METHODS:Rats in the model and control groups were performed digging skull operation after anesthetized. A total of 10 g/L 6-hydroxydopamine or 8 μL physiological saline was injected at the nigral cell bodies of left brain,respectively. There was no treatment in the normal group. MAIN OUTCOME MEASURES:Power of brain 11 mHz infra-slow wave spectrum was examined by encephalofluctuograph technology (EFG),and content of brain dopamine was detected by high performance liquid chromatography (HPLC),in addition,the data of two examinations were analyzed by correlation analysis. RESULTS:Compared with the normal group,content of brain dopamine decreased 26.98% in the left brain,increased 16.92% in the right,and the power of 11 mHz infra-slow wave spectrum decreased 64.69% in the left,decreased 57.67% in the right in the control group. Compared with the normal group,the content of brain dopamine of the model group decreased 83.42% in the left brain,increased 27.48% in the right,and the power of 11 mHz infra-slow wave spectrum decreased 83.39% in the left,decreased 76.62% in the right. There was a positive correlation between content of brain dopamine and 11 mHz infra-slow wave spectrum in each group. (P 0.05-0.01) CONCLUSION:There is a high correlation between the outcomes of the two examinations,namely there is a high correlation between the power of brain 11 mHz infra-slow wave spectrum and the content of brain dopamine,which indicates the corresponding relation between the power of brain 11 mHz infra-slow wave spectrum and the content of dopamine under the conditions that the brain transmitter content lower than or equal to the normal level. Therefore,it is feasible to estimate the brain dopamine content through measuring the power of brain 11 mHz infra-slow wave spectrum.
