Research on Pollen Germination and Pollen Preservation Characteristics of Acer palmatum

The pollen vitality and storage capacity of Acer palmatum were characterized by using in vitro culture. The result showed that the optimal pollen culture time was 24 hours. Sucrose and boric acid affected pollen viability remarkably,and the optimum culture solution was15 % sucrose + 0. 01 % H_3BO_3. Different storage conditions had great effects on the germination of pollen,and pollen viability decreased with the extension of storage time. The pollen viabilities under natural conditions decreased very fast,and reached 0 after 15 days,while those of under the temperature of 4 ℃ decreased slower,and still maintained 28. 1 % and the pollen viabilities under the temperature of-20 ℃ maintained 46. 6 % after 20 days. Therefore,the temperature of 4 ℃ and- 20 ℃ were suitable for storage capacity of A. palmatum such as for pollen stored in the 15 d,the refrigeration and freezing conditions were both possible; If kept for more than 15 days,then frozen condition was more appropriate.
