Effect of carotene beta on the expression of α-SMA and TGFβ_1 in rats with hepatic fibrosis

Objective To study the effects of the carotene beta on the expression of smooth muscle actin alpha(α-SMA) and transforming growth factor-β1(TGF-β1) in rats with hepatic fibrosis.Methods Forty-eight rats was randomly divided into three groups:control group,fibrosis group and interrupt group.The expression of α-SMA and TGF-β1 was showed by immunohistochemistry dyeing,hepatic microstructure was showed by HE staining,hepatic ultramicrostructure was observed by transmission electron microscope(TEM).Results There was significant difference in the average grey degree of α-SMA and TGF-β1 among control group,fibrosis group and interrupt group(P0.01).TEM showed,there were many fatty droplets in hepatic stellate cell(HSC),but no collagen fibers around them in control group;the lipid droplets in HSC were fewer in fibrosis group than these in control group,and amount of the droplets in interfere group between the contrast group to fibrosis group.Conclusion carotene beta can inhibit the expression of α-SMA and TGF-β1,and decrease the hepatic fibrosis of rats by CCl4.Its function may be related with inhibiting the losing of fatty droplets in HSC.
