Culture of vascular endothelial cells from mouse artery

Objective To develope an efficient protocol for the isolation and culture of vascular endothelial cells from mouse aorta.Methods The aorta was removed from mice under an operation microscope,with the periadventitial fat and connective tissue cleared of carefully.The vessel was opened longitudinally and cut into small 1 mm×1 mm cubes.The cubes were placed on six-well plates with the intima side down and covered with a little DMEM containing 20% fetal bovine serum.The cell growth was evidenced by revealing the morphological characteristics and immunological marker VIII factor(or von Willebrand factor,vWF).Results Endothelial cells were found migrate out of the aortic cube after 60 h,forming confluent monolayer of a cobblestone pattern when observed under inverted phase-difference microscope about 12 days later.Immunohistochemical labeling showed the characteristic positive reaction to vWF in the cytoplasm of the new cells.Conclusion A reliable method is described to isolate and propagate vascular endothelial cells from mouse aorta by cultivating small tissue cubes.
