EFFECT OF 4-PHOSP ATE-HYDROXYPIPERAQUINE ON THE ISOLATED AURICAL OF GUINEA PIG

The effect of 4 -Phosphate- lydroxypiperaquine ( PHPQ) on the phsiologic properties in isolated auricle of guinea pig was studied.
Twenty minutes after PHPQ 30 μmol/L was added, the contractility dT/dtmax of the left atria of guinea pig were decreased to 74.4 ± ( SD ) 5.3%(n=6, P0.01) 73,8±4.9% (n=6, P0.01) resp- ectively, the functional refractory period ( FRP ) was prolonged from 139.4±26.7 ms to 214.8±27.8 ms (n=9, P0.01), the duration -intensity curve was shifted remrkably to up-right direction ( n= 8 , P0.01) the epinephrine concentration inducing automaticity was increased from 33.4±6.2μmol/L to 106.9±21.6 μmol/L (n=7, P 0.0l). Furthermore ,30 μmol/L PHPQ could decrease the spontaneous beating rate from 148±16 beat/min to 142±15 beat/min (n=6, P 0.01).
The results suggest that PHPQ might be antagonistic to Na+ in current also influence the utility of Ca2+ . The decrease of automaticity excitability . the prolongation of FRP are thought to be related to its antiarrhythmic action.
