The regulating effect and mechanism of Sini decoction on vascular active substance in renovascular hypertensive rats

Aim To investigate the regulating effect and mechanism of Sini decoction(SND)on the vasoactive substances expression in main target organs and the contents in blood.Methods A total of 24 healthy female Wistar rats were randomly divided into sham operation(SO)group,renovascular hypertension(RH)group and SND treatment(ST)group consisting of 8 cases in each group.The renovascular hypertensive rat models were established according to two-kidney two-clip(2K2C)method.The blood pressure of rats was measured by RBF-2 tail blood pressure meter.The content of nitric oxide(NO)in serum was determined by nitrate reductase assay;and the contents of endothelin(ET),calcium gene related peptide(CGRP)and angiotensinⅡ(AngⅡ)in plasma were determined by radioimmunoassay.The expressions of endothelin receptor A(ETRA),nitric oxide synthesis(NOS),AngⅡ,and CGRP in heart,brain and kidney tissue were determined by immunochemistry assay.Results ① The blood pressure level in ST group was lower than that in RH group(P0.05).② The NO contents in serum and the CGRP and ET in plasma in ST group were higher than those in RH group(P0.05),while AngⅡ had no significant change(P0.05).③ The expression levels of ETRA in heart and kidney in ST group were lower than those in RH group(P0.05),but there was no significant difference in brain(P0.05).The expression level of eNOS in heart in ST group was higher than that in RH group(P0.05),but the expression levels of iNOS in brain and kidney in ST group were significantly lower than those in RH group(P0.05).The expression levels of CGRP in heart,brain and kidney in ST group were markedly lower than those in RH group(P0.05).The expression levels of AngⅡin heart and kidney in ST group were markedly lower than those in RH group(P0.05),while there was no significant difference existed in brain(P0.05).Conclusions SND might regulate the blood pressure in hypertensive rats by adjusting the vascular active substances expression in main target organs and the levels in blood.
