Effect of exendin-4 on the expression of survivin in pancreas in streptozotocin-induced diabetic mice

Aim To investigate the effect of exendin-4 (Ex) on the expression of survivin in pancreas in streptozotocin (STZ)-induced diabetic mice.Methods Balb/c mice (6-weak-old) were divided into three groups: STZ group,Ex group and control group. Blood glucose concentrations,immunocytochemistry and transferase-mediated dUTP nick-end labeling assay (TUNEL) were performed in the present study. Expression of survivin mRNA was detected by real time PCR.Results Blood glucose concentration in Ex group was lower than that in STZ group during the experimental period.At the end of experiment,the percentage of insulin positive area in Ex mice were higher than that of STZ mice (39.40±9.72 vs 25.47±9.26,P0.01).Apoptotic islet cells in Ex group were decreased than those in STZ group. Expression of survivin gene in pancreas was significantly increased at the end of 4th week after STZ administration,and it was higher in Ex group than in STZ group.Conclusion Exendin-4 reduces islet cell apoptosis in STZ-induced diabetic mice,which is partially mediated by up-regulating expression of survivin.
