Effect of Taohongsiwu Decoction on the Proliferation of Hepatic Stellate Cell in Rats with Hepatic Fibrosis

Objective: To observe the impact of Taohongsiwu decoction on the proliferation of hepatic stellate cell( HSC) in rats with hepatic fibrosis,and explore its anti- hepatic fibrosis effect. Methods: The rats were randomly divided into 4 groups as the control group,the model group,the group with low dose of Taohongsiwu decoction and the group with high dose of Taohongsiwu decoction with 10 rats in each group. Rats in all groups except the normal group were injected with CC14 to prepare the rat model of hepatic fibrosis. Corresponding doses of Taohongsiwu decoction water solution were administrated in rats of the low dose group and the high dose group through intragastric administration. Biochemical indexes of hepatic function of rats in each group were determined including serum alanine aminotransferase( GPT),aspartate aminotransferase( GOT),total bilirubin( Bil),total protein( TP) and albumin( Alb). Detection of the proliferation and apoptosis of HSC was also performed and the morphological changes of hepatic tissue were observed. Results: Compared with the model group,rats in the Taohongsiwu decoction group showed obviously lower serum levels of GPT,GOT and Bil( P 0. 05 or P 0. 01),evidently higher levels of TP and ALB( P 0. 05),obviously enhanced apoptosis rate of HSC( P 0. 05 or P 0. 01),and evidently increased percentage of HSC in the G0 / G1 phase. Histomorphology improvement was achieved in the experimental hepatic fibrosis. Conclusions: Taohongsiwu decoction greatly improves the basic biochemical indexes of rats with CCL4 caused hepatic fibrosis and postpones fibrosis development,and its function mechanism may be associated with the acceleration of HSC apoptosis and inhibition of HSC proliferation.
