Comparative Studies of Esterase Isozymes among Triticum, Secale and Triticale

Differences of the esterase isozyme zymograms of endosperms at milk stage among 72 varieties of Triticum,Secale and Triticale were studied with polyacrylamide gel electrophoresis on vertical slab.Differences of esterase zymograms among intra-varietal plants in these three genera were further determined.The marker hands of esterase isozyme for tetraploid wheat and hexaploid wheat all possessed E1-1 E1-2 and E1-3 bands in zone E:.The marker band of T.monococcum appeared between E1-2 and E1-3,while that of Secale was at E1-5.But in some of the Secale cereal varieties,there were three hands with weak activity and their migration rate was similar to the above-mentioned E1-1,E1-2 and E1-3.In Triticale there were five marker bands in zone Ei.There was no difference between hexaploid Triticale (AABBEE) andoctoploid Triticale (AABBDDKE),as well as in the case of tetraploid wheat (AABB) and hexaploid wheat (AABBDD).There was no difference among the various marker bands observed in our experiments.The presence or absence of D genome did not affect the occurrence of the five marker bands.Three of them were similar to those of tetraploid and hexaploid wheat in migration rate,and one of the marker bands was similar to that of Secale.But they were different in activities,and the band E1-4 was a hybrid band.Sampling for determination from the mechanical mixtures of wheat and Secale had only four bands: E1-1,E1-2,E1-3 and E1-5.In some varieties of Triticum,Secale and Triticale,there were differences in zymogram of esterase isozyme in migration rate,activities and band numbers among intra-varietal plants,although no significant differences were seen in morphological and biological characteristics among plants within the same variety.The zymogram differences of esterase isozyme among intra-varietal plants were greater in Secale cereal (about 42.1%),smaller in Triticum (17.6%),and the smallest in Triticale (6.2%).
