Study on the homozygous deletion of multiple tumor suppressor gene in non-small cell lung cancer and its clinical significance

Objective The homozygous deletion of MTS1 gene (p16)of the patients with non small cell lung cancer (NSCLC) was investigated and combined their clinical information to explore the association with the histological type of differentiation and prognosis. Methods 32 surgical samples of Chinese patients with NSCLC were diagnosed by pathology. DNA was extracted from these tissues and the homozygous deletion of p16 gene including exon 1 and 2 were detected with polymerase chain reaction (PCR). The PCR products of p16 exon 2 were analyzed by Southern blotting. Results The homozygous deletion of p16 exon 2 were found in 9 cases of 32 cases. The homozygous deletion rate of p16 exon 2 was 28%. No homozygous deletion of p16 exon 1 were detected in these samples. Conclusion In these NSCLC patients the homozygous deletion of p16 exon 2 may be a major way of the inactivation of p16 gene and affected biological behaviour of lung cancer.
