The Effect of Endothelin 1 on cAMP and cGMP level in Retina of Guinea pig

bjectiveTo investigate the relationship between the balance of microcirculation and metabolism in retina of guinea pig with the level of ET 1 in systemic and retinal blood vessels.MethodsThe cAMP and cGMP level of retinas were measured in 26 guinea pigs after a single intravenous injection of ET 1(Endothelin 1)on 10,20,40 and 80 minutes as well as 7 normal controls by radioimmunoassay.ResultsThe cAMP and cGMP level and the ratio of cAMP/cGMP were decreased compared with the normal controls after the injection,particularly on minutes 20,the cAMP level of retinas was the lowest(P0.001).On minutes 40,the cGMP level as well as the ratio of cAMP/cGMP(P0.01) declined to the lowest(P0.05),and then they gradually recovered up to the lower of the normal level respectively (P0.05).ConclusionThe balance of microcirculation and metabolism in retina of the guinea pig is related to the level of ET 1 in systemic and retinal blood vessels.ET 1 may play an important role in regulating the normal metabolism of the retinal tissue.
