Protective effect of gypenosides on acute incomplete cerebralischemia in rabbits

The protective effect of gypenosides (GP) extracted from Gynostemma pentaphyllum (Thunb.) Mak. on acute incomplete cerebral ischemia in rabbits was studied. This ischemia-model was produced by bilateral carotid artery occlusion coupled with hemorrhagic hypotension. 30 rabbits were randomly divided into 3 experimental groups, 10 in each group. Group 1: sham operation; Group 2: ischemia for 60 min; Group 3: GP and ischemia 60 min. GP (50 mg·kg-1) was slowly injected iv 30 min before ischemia. In the ischemia group, normal saline was injected iv in the same volume as GP. By the endof 60 min of induced ischemia, GP significantly improved the brain electrical activity and histological and ultrastructural ischemia changes and suppressed the rise of lactate dehydrogenase (LDH) and creatine phosphokinase (CPK) release in the cerebral venous blood. These results indicate that GP has the protective effect on acute cerebral ischemia injury.
