STUDIES ON MABINLIN, THE SWEET PROTEIN FROM SEEDS OF CAPPARIS MASAIKAI Ⅳ. STABILITY AND DENATURATION

After a scries of heating denatnration experiments, we found the sweetness of Ma I was irreversibly lost in incubating at 80℃ for 30 minutes, the α-helix of Ma I also disappeared totally and the PAGE behavior changed. But the sweetness of Ma Ⅱ wasn't lost at 80℃ for 48 h incubation, and its PAGE behavior didn't change. In denaturant tests, a bathochromic shift of Try fluorescence emission pack of Mabinlin happened along with the increasing of the concentration of denaturants untill the clenaturation has been complete. By means of this indicator, we also found that in acidic condition Mabinlin was more stable, that 3-5 M Gdn-HCl was strong enough to denature Mabinlin, but even 8 M urea had a slight effect. This was a similar result as that of transverse urea-gradient electrophoresis. If the denaturants were removed, the sweetness of Ma I might recover, the secondary structure and the PAGE behavior of Ma I only altered slightly. There was a dimer band of Ma I in SDS-PAGE analysis. But after Ma I was thermodenatured, or denatured by DTE and Gdn-HCl or carboxymc-thytated on its SH groups, etc., the dimer band disappeared in the gel as the loss of the sweetness of Ma I. In case of Ma Ⅱ, the dimer band never appeared when any method mentioned above was used. The formation of dimer band might be due to the specific structure fo Ma I that related to its sweetness.
