HPLC Determination of Telmisartan in Human Plasma with Fluorescence Detection

Objective:To develop an HPLC method for the determination of telmisartan in human plasma.Meth- ods:1 mL of plasma sample was mixed with 100 μL of 15% EDTA-K_2 solution,then extracted with 4 mL of ethyl acetate.The analyte was separated on a Diamonsil C_(18)(150 mm×4.6 mm,5μm)analytical column with fluores- cence detection at λ_(es) 305 nm and λ_(ex)m 365 nm.The mobile phase consisted of acetonitrile-methanol-0.05 mol· L~(-1)sodium dihydrophosphate solution (4:3:3,pH 6.7)and the flow rate was 1 mL·min~(-1).Doxazosin mesilate was used as internal standard.Results:The linear range of the plasma concentration of telmisartan was 2-500 ng· mL~(-1)and the limit of quantitative was 2 ng·mL~(-1).The relative recoveries of telmisartan were 100.6%-105.3%. The RSDs of intra-day and inter-day were less than 7% and 12%,respectively.Conclusions:This method is simple,selective and sensitive.It' s suitable for the pharmacokinetic research of telmisartan in human plasma.
