Optimization on extraction technology for Eucommia ulmoides by single-factor experiment combined with response surface methodology

Objective:To approach an optimal extraction process with the content of pinoresinol diglucoside(PDG) as an index for Eucommia ulmoides,and provide a basis for the in vivo pharmacokinetic studies in rats after oral administration of the Eucommia ulmoides extract.Methods:The single factor experiment was utilized combined with response surface methodology to optimize the extraction process of Eucommia ulmoides.On the basis of single factor experiments,different optimum conditions for the extraction of PDG from Eucommia ulmoides were selected,and the method of response surface analysis with 3 factors and 3 levels was adopted according to the Box-Benhnken center-united experimental design principles.Results:The results showed that the optimum conditions for extraction of PDG from Eucommia ulmoides were as follows:Eucommia ulmoides was extracted using 26% of ethanol solution with the ratio of material to solvent at 1∶ 13 for 25 min by ultra sonification,under which the extraction predictive value of PDG was 3.2809 mg·g-1.Conclusion:Compared with the method described in Chinese Pharmacopoeia(version 2010),this method has advantages of higher extraction yield,lower energy consumption,and shorter extraction time,which will lay a good foundation for the subsequent in vivo pharmacokinetic study of Eucommia ulmoides.
