Effect of KunNingAn on estrogen receptor and mRNA in hypothalamus of perimenopause syndromes rats

Objective To explore the Effect of KunNingAn concentrating pill on the whole process of estrogen biological effect,which including estrogen(E2)-estrogen receptor(ER)-estrogen receptor mRNA(ER mRNA),thereby to find the mechanisms that KunNingAn concentrating pill play an influential role in prevention and treatment of perimenopause syndromes(PMS).Methods After animal models of PMS were built up,rats were divided into six groups:younger control group(group A),model group(group B),low dosage and high dosage of kunningan(group C and D),GengNianAn concentrating pill control group(group E),Premarin control group(group F).Changes of ER,ER mRNA in Hypothal tissue were analyzed by immunobiochemistry and in situ hybridization technologies,level of E2,GnRH,FSH,LH in blood were detected by radioimmunoassay.Results Blood concentrations of E2 in group B was lower than group A while GnRH,FSH,LH were rather high,and E2,GnRH,FSH,LH of group C,D,E showed no significant difference compared to group B(P0.05),significant difference of E2 was found in group F compared to group B(P0.01).The concentration of ER and ER mRNA in hypothal tissues were found lower in group A than group B.ER and ER mRNA of group C,D,E were all found increased.The most increased changes was seen in group D.It is significant difference between groupA and group B.While no significant difference was found in group F compared to group B(P0.05).Conclusion By increasing the expression of ER mRNA in rats′Hypothal tissue,so as to increase the production of ER,KunNingAn concentrating pill can play its role by increasing ER concentration in target tissues and enhance the biologic effect of E2.
