Effects of Berberine on Macrophage Function in Hypoxic Injury of H9c2 Cardiomyocytes

Objective To investigate effects of berberine(BBR)on macrophage function in hypoxic injury of H9 c2 cardiomyocytes in mice.Methods Rat bone marrow-derived macrophages(BMDM)as well as co-cultured BMDM and rat H9 c2 cardiomyocytes were treated with BBR for 24 h to determine the toxicity of the two groups.At the same time,high mobility group box-1 protein(HMGB1),inflammatory factors IL-6 and IL-1β in BMDM macrophage treated by BBR for 24 h were measured.Results BMDM culture and combined culture were not toxic by 10μmol/L BBR management,which could reduce levels of HMGB1 and inflammatory factors.In combined culture,10μmol/L BBR management could control macrophage and reduce apoptosis in hypoxic cells.Conclusion Hypoxic injury of H9 c2 cardiomyocytes in mice with berberine treatment has a good regulation of macrophages and pro-inflammatory functions,and then achieve the role of repairing defective cardiomyocytes.
