Simultaneous Determination of Four Alkaloids in Radix et Rhizoma Sophorae Tonkinensis by HPLC and Cluster Analysis

Objective To establish an HPLC method for simultaneous determination of matrin, oxymatrine, sophocarpine and oxysophocarpine in Radix et Rhizoma Sophorae tonkinensis. Methods The Agilent Zorbax SB C18 column( 150 mm × 4. 6 mm,5 μm) was adopted with methanol( A)-0. 2% phosphoric acid solution( B)( adjusted p H = 5. 0 by triethylamine) as mobile phase in gradient elution. The flow rate was 1 m L/min and detection wavelength was 215 nm,the column temperature was 30 ℃. Results The linear ranges of matrin, oxymatrine, sophocarpine and oxysophocarpine were 31. 40-2 910. 47 μg/m L, 28. 70-164. 53 μg/m L, 50. 16-813. 43 μg/m L,71. 43-4 300. 00 μg/m L( r ≥ 0. 999 0), the average percentages of recovery were in the range of 100. 41% to 101. 23% with the RSD between 1. 82 % to 2. 16 %( n = 6). Conclusion This method is simple, accurate and repeatable. The content of 4 alkaloids in different habitats is different, and the content of four alkaloids in geo-authentic habitats of Radix et Rhizoma Sophorae tonkinensis was relatively high.
