Detecting minimal residual disease of leukemia with hypermethylation of the calcitonin gene as molecular genetic marker

Objective:To explore whether detecting minimal residual disease of leukemia with hyper-methylation of the calcitonin gene as molecular genetic marker of leukemic clone to predict the prognosis.Methods: Polymerase chain reaction in combination with digestion of DNA with Hpa Ⅱ was used to ex-amine the methylation patterns of the calcitonin gene in 29 cases with acute leukemia and 8 cases withtransformation of chronic myeloid leukemia. By using polymerase chain reaction, minimal residual diseasewas longitudinally detected in patients which were positive for hypermethylation of the calcitonin gene withhypermethylation of the calcitonin gene as molecular genetic marker. Results: All of patients with leuke-mia had minimal residual disease after complete remission. Bone marrow relapse was occurred soon whenminimal residual disease persisted or reappeared. It may predict bone marrow relapse ahead of two toeleven months. The patients which got early negative for minimal residual disease and were persistent neg-ative may acquire prolonging survival. Conclusion: Minimal residual disease of leukemia may be moni-tored by using polymerase chain reaction with hypermethlation of the calcitonin gene as molecular geneticmarker for leukemic clone. It may prove useful in predicting prognosis of leukemia.
