Prokaryotic Expression and Immunogenicity of the CT703 Protein from Chlamydia Trachomatis

Objective To construct the prokaryotic expression plasmid pGEX/CT703,express CT703 protein and study its immunogenicity in mice.Methods The entire DNA fragment of CT703 gene was obtained from the Chlamydia Trachomatis(CT) serovar L2 genome by RT-PCR,and was subcloned into the pGEX plasmid.The recombinant plasmid was transformed into competent E.coli BL21 to express GST-CT703 fusion proteins after identification by restriction enzyme digestion and sequence analysis.The GST-CT703 fusion proteins were used to immunize BALB/c mice.ELISA and Western blot were carried out to identify the immunogenicity and immunoreactivity.Results The recombinant expression plasmid pGEX/CT703 had been constructed successfully,which was demonstrated by restriction enzyme digestion and sequence analysis.The expressed fusion protein induced by IPTG had a relative molecular mass of 81KD.Western blot assay demonstrated the antiserum bound specifically CT703 protein,and ELISA result showed the specific antibody titer of CT703 reached 1:320 00.Conclusion CT703 protein can be expressed successfully in E.coli BL21 and the specific anti-CT703 antibody is induced in BALB/c mice.
