Determination of rutin and quercetin in Gynostemma pentaphyllum (Thunb.) Makino. by RP-HPLC

AIM:To determine main flavoniods rutin and quercetin in the Gynostemma pentaphyllum(Thunb.)Makino.by RP-HPLC.METHODS:The separation was performed on TURNER Kromasil TMC_ 18 column(250 mm× 4.6 mm,5 μm),the mobile phase consisted of 0.4% phosphoric acid-methanol-acetonitrile,with the flow rate of 0.5 mL/min at 40 ℃,the wavelength for measurement was at 360 nm.RESULTS:The calibration curves were linear in the range of 0.37-3.70 μg for rutin(r= 0.999 9)and quercetin(r= 0.999 9),respectively.The average recoveries of compound rutin was 99.6%(RSD? ?.07%),and that of compound quercetin was 97.2%(RSD? ?.01%),respectively.CONCLUSION:This method is accurate and reproducible.
