Objective:To explore the effect factors of onset and severity of congenital megacolon, provide a basis for prevention and treatment of congenital megacolon.Methods:253 couples of parents whose infants were diagnosed with congenital megacolon definitely received face-to-face questionnaire investigation, the contents included 26 indexes, such as the general conditions of infants and their parents, the physiological and psychological situations of their mothers before and during pregnancy, partial environmental factors and family history of related severe diseases.253 children without congenital gastrointestinal tract anomalies during the same period were selected as control group. All the data were analysed with SAS9.0 software by Chi-square test, nonparametric test, single factor and multi-factor non-conditional logistic regression. Results:There was significant difference in the incidence of menstrual disorder at six months before pregnancy, the incidence of maternal abdominal pain during the first trimester of pregnancy, the rate of active or passive smoking, the rate of viral infection, the contact rate of toxic substances such as paint during pregnancy, the proportions of cases with family histories of malignant tumors and congenital deformities between case group and control group(P0.05).The increase of maternal age was related to the aggravation of congenital megacolon. The results of multivariate analysis showed that the risk factors of congenital megacolon with OR values from high to low were viral infection, maternal abdominal pain during the first trimester of pregnancy, menstrual disorder at six months before pregnancy, active or passive smoking during the first trimester of pregnancy and toxic substances contact such as paint during pregnancy. Conclusion:Genetic factor, environmental factor and biological factor are related to the occurrence of congenital megacolon.Active or passive smoking during the first trimester of pregnancy and viral infection may participate in the onset of congenital megacolon and affect the severity of the disease by various ways.
