The Transfection to BMSCs with Eukaryotic Expression Plasmid of pBudCE4.1_Cx43 And the Evaluation of Their Expression Product and Function

Objective To study the expression and function of rat bone marrow stromal cells (BMSCs)after cells were transfected with eukaryotic expression plasmid of pBudCE4.1_Cx43 in vitro. Methods The rat BMSCs were transfected with eukaryotic expression plasmid of pBudCE4.1_Cx43 together with lipofectamin2000,The rat BMSCs were transfected with eukaryotic expression plasmid of pBudCE4.1_ Cx43 together with lipofectamin2000,then Cx43mRAN was measured by hemiquantitative RT-PCR,Cx43 protein was measured by immunocytoehemical,cell gap junction communication was measured by scrape- loading and dye transfer(SLDT).Results There is no significant diversity in morphology,proliferate capac- ity between transgenic cells and BMSCs,while the RT-PCR measur the expression of Cx43mRNA in BMSCs after transfection were higher than the control group,Map analysis indicated the Cx43 protein expression in BMSCs after transfection were higher than control group.Both Cx43 mRNA and protein,the difference be- tween the two group was significant(P0.01).In the experiment of SLDT,the fluorescence intensity of transgenic BMSCs transferred was higher than control group,the difference between them was significant (P0.01).Conclusion Transfected BMSCs could effectively express the active Cx43 and produce biolog- ical efficiency.It will offer the ideal genetically engineered cell for cellularity cardiomyoplasty.
