Histopathological Changes of the Extracellular Matrix Caused by Genotype Ⅶ and Ⅳ Newcastle Disease Viruses of the Spleen in Chicken

Histopathological changes of extracellular matrix(ECM) caused by genotype Ⅶ Newcastle disease virus(NDV) JS5/05 and genotype Ⅳ NDV Herts/33 in chicken spleens were systematically analyzed and compared.Pathological changes of the spleens were examined and ECM histology was investigated using ECM-specific staining,including Masson′s trichrome and Picrosirius red staining.In addition,distribution of a key ECM component(collagen Ⅰ) in the spleens from virus-infected birds was determined by immunohistochemistry(IHC).The results demonstrated that JS5/05 caused much more severe pathological changes in the spleen characterized with remarkable necrosis compared to Herts/33.Moreover,ECM-specific staining revealed that JS5/05 induced a significant decrease in the intensity of ECM components and also a remarkable disruption of the integrity of collagen fibers in the spleen compared to Herts/33.Additionally,IHC demonstrated that decreased intensity of collagen Ⅰ and breakdown of collagen Ⅰ network were observed in JS5/05-infected spleens.It′s suggested that ECM breakdown in chicken spleens was a critical pathological event in the process of genotype Ⅶ NDV infection and the histologic changes of the ECM might be associated with the immunopathology phenotype of genotype Ⅶ NDV.
