Establishment of Model of Traumatic Brain Injury by an Improved Method in Rats

Objective To establish traumatic brain injury (TBI) model with different severity in degree. Methods The right cerebra in 30 Wistar rats, who were averagely into 3 groups, were injured respective by 20 g·cm, 50 g·cm and 100 g·cm impact strength released from the weight drop equipment improved by Weiss Yong. The changes in the systemic reaction, pathology of cortexes and hippocampi, and the behavior were observed after TBI in all the rats. Results The only mild contusion of cortexes was produced by 20 g·cm strength. The middle contusion of cortex was observed and the hippocampal structure is normal in 50 g·cm strength group. The strength of 100 g·cm led to significant contusion of cortexes and hippocampi. The latency of avoidance reaction in 20 g·cm group was significantly longer only 1 day after TBI than that in the control group (P0.01). The latency of avoidance reaction in 50 g·cm group was significantly longer 1, 3 and 7 days after TBI than those in 20 g·cm group and control group (P0.05 or P0.01). The latency of avoidance reaction in 100 g·cm group was significant longer than those in the other 3 groups (P0.05 or P0.01). Conclusions The traumatic brain injury models with mild, moderate and severe brain injury can be well established by 20 g·cm, 50 g·cm and 100 g·cm impact strength released from weight drop equipment improved by Weiss Young.
