CHANGE 0F PULMONARY BETA─ADRENERGIC RECEPTOR AND ITS RELATI0N TO METABOLISM 0F MEMBRANOUS PHOSPHOLIPIDS DURING ENDOTOXIN─INDUCED RAT LUNG INJURY

Change of beta─adrenergic receptor(β─AR) in lung tissue during endotoxin─induced ratacute lung injury was observed with radioligand binding assy.The lipid fluidity andphospholipids content of the cellular membrane of lung tissue were measured with fluorescencepolanzation and high performance liquid chromatography respectively.The results showed that :(1) 4 hours after the endotoxin injection,there was a significant decrease in the maximal bindingcapacity of β─AR by 47% as compared with the control group;(2) endotoxin could markedly de-crease the lipid fluidity and phospholipids content of lung cell membrane,meanwhile there waselevated activity of phospholipase A2(PLA2) in the tissue of rat lungs.The data suggest that:(1)the down regulation of β─AR,that can diminish the function mediated by β-AR, plays a role inthe pathogenesis of endotoxin-induced acute rat lung injury;(2)the activation of PLA2 can bean important factor reducing the phospholipids content of cell membrane and thus leading to thedecrease of membranous lipid fluidity,resulting in reduction of the lateral diffusion and rotatorymovement of β-AR and decrease in chance of β-AR to bind with the ligands.As a result ofthese changes,down regulation ofβ-AR appears.
