Study on simultaneous detection of 7 tetracyclines residues in aquatic products by high performance liquid chromatography

A HPLC method for simultaneous determination of residues of tetracycline,chlortetracycline,oxytetracycline,doxycycline,demeclocycline,methacycline,and minocycline in aquatic products was developed.In the method,the analysis column was reversed-phase column of Inertsil C8-3(5μm,250mm×4.0mm i.d.),the extraction solution was EDTA-McIlvaine buffer solution(pH4.0),the clean-up column was HLB SPE column,the mobile phase was methanol-acetonitrile 0.01mol/L trifluoroacetic acid solution with gradient elution,the flow rate was 1.5mL/min,the detection wavelength was 350nm,and the injection volume was 100μL.The limit of detection was between 1.5μg/kg and 5.0μg/kg,and the limit of quantitation was 50μg/kg.The linear plots were obtained between 50 and 1000μg/kg.Overall recoveries were between 75.2% and 100.6% with RSD(n=10) values between 2.19% and 10.56%.The results showed that the method was sensitive,accurate,convenient and quick,which can be used to detect simultaneously the 7 tetracyclines residues in aquatic products.
