Extraction of Enriched Icarrin from Epimedium Crude Extracts

OBJECTIVE:To separate icarrin from crude extracts of epimedium by extraction technology.METHODS:The crude extracts (the content of icarrin at 2.01%)were pretreated.The ideal extraction solvent was selected with the consideration of the category of solvents,the influencing factors of one class extraction and the observation of all extraction methods.RESULTS:The extraction yields of methyl acetate,ethyl acetate and butyl acetate extracting solvents in single-stage extraction were 33.55%,29.53%and 17.18%,respectively.The contents of icariin in extracts were 8.89%,10.4%and 6.99%,respectively.When extraction temperature,basic material concentration,phase ratio and extraction time were 30℃,3mg/mL-1,1∶1 and 10 min,respectively,the content of icariin was the highest in single-stage extraction with acetic ether as extracting solvent.Its extraction yield of icariin was about 30%.Both the extraction ratios of countercurrent multi-stage extraction and cross current multi-stage extraction were higher than single-stage extraction.The extraction rate of icarrin could reach as high as 90.24%by countercurrent multi-stage extraction with stages and phase ratio at 3 and 4∶1 respectively,and the content of icarrin was 20.5%.These results were consistent with the theoretic ones.CONCLUSION:Acetic ether was chosen as the extraction solvent for it is low-toxic,economical,less contaminative and can be reclaimed.This extraction technology can provide technological support for industrial production.
