Recent advances in pharmacokinetic-pharmacodynamic modeling and its applications in drug research and development

Pharmacokinetic-pharmacodynamic(PK-PD) modeling is a powerful tool which links pharmacokinetics and pharmacodynamics and describes the inter-relationship among dosage or concentration,time and response,thereby gaining ever-increasing applications in the drug development process and dosage optimization.According to the manner in which the measured pharmacokinetic and pharmacodynamic data are related to each other,PK-PD models can be classified into four kinds of basic models.In recent years,the applications of neural networks technology and optimal design theory in PK-PD study promote the development of PK-PD models with multicomponents and multitargets.Finally,this paper presents the applications of PK-PD modeling in the preclinical and clinical stages of drug research and development intensively as well as its current hot spot and tendency.
