Polymorphism of human cytomegalovirus UL138 gene in low-passage clinical isolates

Objective: To investigate the polymorphism of human cytomegalovirus UL 138 gene in low passage clinical isolates and to find the relationship between the polymorphism and different pathogenesis of congenital HCMV infection. Methods:Polymerase chain reaction (PCR) was performed to amplify the entire HCMV UL 138 gene region of 30 clinical isolates, which had been proven containing detectable HCMV DNA by using FQ PCR.The UL138 gene in 16 clinical isolates were sequenced directly and the sequence data were analyzed.Results:Twenty isolates were amplified successfully.Open reading frome of HCMV UL138 in 16 low passage clinical isolates was highly conserved, regardless of their polymorphism. The phylogenetic tree demonstrated that congenital megacolon isolates existed in genotype 2a only and microcephaly isolates existed in genotype 2b exclusively except for strain 20M. Conclusion:HCMV UL138 gene in clinical isolates was highly conserved,regardless of their polymorphism.
