miR26a Suppresses Proliferation of Hep G2 Cells by Targeting EZH2

Objective To investigate the effect of miR26 a on proliferation of human hepatocarcinoma Hep G2 cells and its relationship with EZH2.MethodsSynthetic miR26 a mimic and its negative control were transfected into Hep G2 cells by liposome method. We divided cells into threegroups:mock group,control group and mi R26 a mimic group. After transfection,the expression of miR26 a at m RNA level was detected by quantita-tive real-time PCR. The effect of miR26 a on the proliferation of Hep G2 cells was analyzed by MTT,and the percentage of apoptosis was detected byflow cytometric analysis. The target relationship between miR26 a and EZH2 was identified by Dual-Luciferase Assay System. Expression of EZH2 was analyzed by Western blot.ResultsMTT assay showed that miR26 a overexpression significantly inhibited the proliferation of Hep G2 cells. Inaddation,the flow cytometric results showed that miR26 a could promote the apoptosis of Hep G2 cells. The target relationship between miR26 a andEZH2 was identified by Dual-Luciferase Assay System and miR26 a down-regulate the expression of EZH2.ConclusionThe mi R26 a overpressioncould inhibit Hep G2 cells proliferation through targeting EZH2.
