Effect of Total Coumarins of Cortex Fraxini on Acute Gouty Arthritis and Uric Acid Metabolism

OBJECTIVE To investigate the effects of total coumarins of Cortex Fraxini(TCCF) on acute gouty arthritis and uric acid metabolism.METHODS Effects of TCCF orally on edema degree of hind paw was investigated in rats with edema of hind paw induced by injection of sodium urate.Effects of TCCF orally on edema degree of joint,joint liquid,joint synovial histomorphology were investigated in rabbits with acute arthritis of hind knee joint induced by injection of sodium urate.Effects of TCCF orally on serum uric acid were investigated in normal mice and hyperuricemia mice induced by hypoxan-thine.RESULTS TCCF at dose of 40,80,160 and 320 mg·kg~(-1) significantly reduced edema degree of hind paw by sodium urate in rats.Also TCCF at dose of 20,40,80 and 160 mg·kg~(-1) remarkably decreased edema degree of knee joint,knee joint liquid and mended knee joint synovial histomorphology in rabbits with acute arthritis by sodium urate.Moreover TCCF at dose of 50,100,200 and 400 mg·kg~(-1) significantly lowered the uric acid in normal mice and hyperuricemia mice induced by hypoxan-thine.CONCLUSION TCCF shows a remarkable improvement effect on acute gouty arthritis and promotes the elimination of uric acid.
