Effects of Triptolide on Human Colon Carcinoma Cells SW-480 and HT-29 in Vitro and in Vivo

OBJECTIVE To study the in vitro and in vivo effects of triptolide(TPL) on human colon carcinoma cells of SW-480 and HT-29.METHODS The effect of TPL on the growth and apoptosis of SW-480 and HT-29 cells were studied by MTT assay,DNA fragmentation and Annexin-V flow cytometry analysis.In vivo,the effect of TPL on human colon carcinoma were studied by establishing the Xenografts of human colon carcinoma in nude mice.RESULTS In vitro,10 g·L-1 TPL significantly inhibited the proliferation of human colon carcinoma cells.However,40 g·L-1 TPL induced the apoptosis of the two cells with the apoptosis rates of 22.2% and 27.3%.In vivo,0.4 mg·kg-1 TPL every other day inhibited the growth of colon carcinoma by 94.8% and 93.7%.CONCLUSION TPL have significant anticancer activities on human colon carcinoma in vitro and in vivo.
