Reversion of Malignant Phenotypes of Human Lung Cancer Cells and Suppression of Its Invasive Ability by Cinnamic Acid

Highly metastatic human lung cancer cell line (PGCL3) was treated with 1-5mmol/L cinnamic acid in vitro. As a result,the PGCL3 cells had their phenotype reversed to their original normal cells in terms of morphology,proliferation rate, mitotic index, colony-forming ability in semisolid agar culture and Con A agglutination reaction. These results showed that cinnamic acid could inhibit proliferation and enhance differentiation of PGCL3 cells. We also found that cinnamic acid treatment could result in significant inhibition of adhesion of cells to laminin substrate, migration ability through the polycarbonate filter of Boyden Chamber,and invasive ability through matrigel as well. In conclusion,cinnamic acid could induce differentiation of PGCL3 cells accompanied by a decline of their invasive and metastatic abilities.
